Electro Mobility Shift Essay (EMSA) A method for detecting DNA-Protein interactions

We preform Quantitative EMSA in order to determine the Association / Dissociation constants of p53 to various DNA sequences of interest.

The EMSA method is a very high sensitivity method. Very low concentration of radioactively labeled DNA can be used and still detect weak bands of protein/DNA complexes. In addition, EMSA is the only method in which the various oligomeric forms of protein /DNA complexes are visible on the gel and can be quantified separately. The analysis of each bound band is computationally achieved using equations based on the probability of observing each bound species.
